Three frequent mutations have been associated with these diseases: JAK2V617F, JAK2 exon 12 mutations and MPL mutations. However, there are multiple causes of dysregulated JAK signaling.

Other mechanisms of dysregulated JAK signaling include excess cytokines, increased JAK1 signaling, loss of negative regulation of JAKs and undiscovered mutations.
